Overview

**MK-2118 + pembrolizumab** is a combination investigational therapy composed of MK-2118, a noncyclic dinucleotide small molecule agonist of the stimulator of interferon genes (STING) pathway, and pembrolizumab (Keytruda), a monoclonal antibody checkpoint inhibitor targeting programmed cell death protein 1 (PD-1). MK-2118 is designed to activate the innate immune response via induction of type I interferon and proinflammatory cytokines through STING pathway activation. Pembrolizumab enhances the immune system’s ability to recognize and attack tumor cells by blocking PD-1, thereby preventing its interaction with PD-L1/L2 and reversing T-cell suppression. The combination has been evaluated in a Phase 1 trial for the treatment of advanced/metastatic solid tumors and lymphomas using intratumoral or subcutaneous MK-2118 administration in combination with intravenous pembrolizumab. This approach aims to synergistically increase anti-tumor immunity, potentially overcoming resistance to checkpoint inhibitors[1][2][3].
