MK-2295 (also known as NGD-8243) is a small molecule antagonist of the transient receptor potential vanilloid 1 (TRPV1), also referred to as the vanilloid receptor 1 (VR1). It was developed as a novel analgesic agent for the treatment of pain, including chronic pathological pain and postoperative pain. MK-2295 works by blocking TRPV1 channels, which are involved in sensing noxious heat and inflammatory agents; this mechanism interrupts pain signaling at the level of sensory neurons. The drug was originally discovered by Neurogen Corporation and further developed in collaboration with Merck & Co., with Ligand Pharmaceuticals also associated with its development. Clinical trials reached up to phase 2 before development was discontinued due to side effects typical of TRPV1 antagonists, such as altered thermoregulation and risk of accidental burns[1][2][3][5][7][8].
