MK-2637 is an investigational small molecule drug developed by Merck & Co. It functions as a glycine transporter 1 (GlyT1) inhibitor, a mechanism that increases synaptic glycine levels and thereby potentiates NMDA receptor activity in the brain. GlyT1 inhibitors are being explored for their potential to treat neuropsychiatric disorders such as schizophrenia, based on the hypothesis that enhancing NMDA receptor function may alleviate symptoms related to glutamatergic dysfunction. MK-2637 has been used in clinical research both as a pharmacological agent and as a tool compound for PET imaging studies to assess GlyT1 occupancy and central nervous system target engagement[3][8]. Clinical trials have evaluated its effects on cerebral cortex excitability using transcranial magnetic stimulation (TMS) and quantitative EEG readouts[8].
