Overview

MK-4166 is a humanized IgG1 agonist monoclonal antibody that targets the glucocorticoid-induced tumor necrosis factor receptor (GITR), also known as tumor necrosis factor receptor superfamily member 18 (TNFRSF18). GITR is expressed on several types of immune cells, including T-cells. By binding to and activating GITR, MK-4166 enhances T-cell proliferation and reduces regulatory T cell-mediated suppression, thereby promoting antitumor immune responses. The drug has been evaluated as monotherapy and in combination with pembrolizumab in patients with advanced solid tumors and melanoma. In clinical studies, it demonstrated tolerability at high doses and showed promising response rates particularly in immune checkpoint inhibitor-naïve melanoma patients[1][2][3][4][5][6].
