Overview

MK-4464 + pembrolizumab is an investigational combination therapy for advanced or metastatic solid tumors, currently in Phase 1 clinical trials. MK-4464 is an antibody-based trifunctional molecule (TriNKET, developed by Dragonfly Pharmaceuticals) designed for immunotherapy in cancer. It binds to CEACAM5 on cancer cells and to NKG2D and CD16A on NK cells, thereby recruiting immune cells into tumors and directly activating the killing of cancer cells; it also engages cytotoxic T cells. Pembrolizumab is a programmed death-1 (PD-1) immune checkpoint inhibitor that enhances the immune response against cancer by blocking a pathway tumors use to evade immune destruction. Their combination aims to synergistically improve anti-tumor immune responses by both directly activating immune effector cells (via MK-4464) and removing immune suppression (via pembrolizumab)[2][1][3][5].
