Overview

MK-4618 + amlodipine is a combination therapy investigated for the treatment of hypertension. MK-4618 is a soluble guanylate cyclase (sGC) stimulator developed by Merck & Co. (MSD). It works by directly stimulating the sGC enzyme and sensitizing it to endogenous nitric oxide (NO), which increases the production of cyclic guanosine monophosphate (cGMP) and leads to vasodilation. Amlodipine is a dihydropyridine calcium channel blocker that inhibits the influx of calcium ions into vascular smooth muscle and cardiac muscle, resulting in peripheral vasodilation. The combination was intended to provide synergistic blood pressure lowering effects by targeting two distinct vasodilatory pathways. Clinical development for this combination in hypertension reached Phase 2 but appears to have been discontinued.
