Overview

MK-4830A is a combination drug consisting of the investigational monoclonal antibody MK-4830 and pembrolizumab (KEYTRUDA). MK-4830 is a first-in-class, fully human IgG4 monoclonal antibody that targets the inhibitory immune checkpoint receptor immunoglobulin-like transcript 4 (ILT4), also known as LILRB2. By blocking ILT4 on myeloid cells in the tumor microenvironment, it aims to relieve myelosuppression and enhance T cell function. Pembrolizumab is an approved anti–PD–1 therapy that blocks the programmed death receptor 1 pathway to promote antitumor immune responses. The combination is being developed for advanced solid tumors and has shown preliminary evidence of antitumor activity in heavily pretreated patients, including those who have progressed on prior anti–PD–1 therapies[3][5][6][8].
