Overview

MK-6186 is a novel small molecule non-nucleoside reverse transcriptase inhibitor (NNRTI) developed for the treatment of HIV infection. It demonstrates subnanomolar potency against wild-type HIV as well as the two most prevalent NNRTI-resistant reverse transcriptase mutants (K103N and Y181C). In phase I clinical trials in HIV-infected patients, MK-6186 showed robust antiviral activity with maximal effect at a dose of 40 mg daily. However, resistance development was observed in some cases due to outgrowth of minority variants such as V106A. The drug was generally well tolerated but its development did not progress beyond early clinical studies[1][6].
