MK-7288 is a novel small molecule drug developed as a histamine H3 receptor inverse agonist. Its primary mechanism of action is to increase wakefulness by inhibiting the activity of the histamine H3 receptor, which plays a key role in regulating the sleep-wake cycle. The drug was investigated for its potential to treat excessive daytime sleepiness (EDS) in patients with obstructive sleep apnea (OSA). In clinical studies, MK-7288 significantly prolonged maintenance of wakefulness test (MWT) sleep latency and improved driving performance in simulated tests compared to placebo. However, it was associated with higher rates of insomnia relative to modafinil and placebo. Development has not progressed beyond Phase 1 trials, and there are no approved indications for this compound[3][4][5][6].
