Overview

MK-8825 is a **potent, selective, orally bioavailable antagonist of the calcitonin gene-related peptide (CGRP) receptor**. It binds human and rat CGRP receptors with high affinity (Ki for rat CGRP ≈ 17 nM). MK-8825 blocks CGRP-induced signaling and inhibits capsaicin-evoked dermal blood flow in animal models, showing significant pharmacodynamic activity. It is structurally related to, but distinct from, other CGRP receptor antagonists such as MK-3207. The drug has been investigated primarily for the **treatment of migraine**, exploiting the role of CGRP in migraine pathophysiology. It is developed by Merck & Co. and has demonstrated efficacy in preclinical models of migraine, including attenuation of trigeminal pain and inhibition of migraine-relevant vasodilatory responses[3][7][11][15].
