Overview

MK2 shRNA is a short hairpin RNA (shRNA) therapeutic candidate designed to knock down the expression of MAPK-activated protein kinase 2 (MK2), a stress-activated kinase downstream of p38 MAPK. In the context of head and neck squamous cell carcinoma (HNSCC), MK2 signaling is associated with poor survival and promotes epithelial-to-mesenchymal transition (EMT), cell migration, and invasion. By utilizing a lentiviral delivery system to silence the MK2 gene, this approach aims to suppress tumor motility and enhance radiosensitivity. Preclinical studies have demonstrated that MK2 knockdown leads to increased expression of cell adhesion molecules like E-cadherin and decreased expression of matrix metalloproteinases and EMT markers such as Snail, effectively shifting cancer cells toward a more epithelial phenotype.
