**MKC1106-PP** is a MannKind investigational multicomponent therapeutic cancer-vaccine regimen for antigen-positive advanced solid tumors. It combines the recombinant DNA plasmid **pPRA-PSM**, which encodes PRAME- and PSMA-derived antigen fragments and epitopes, with the synthetic HLA-A*0201-restricted peptide boosters **E-PRA** and **E-PSM**. The regimen was delivered by ultrasound-guided intralymph-node DNA priming followed by peptide boosting to induce PRAME- and PSMA-specific cytotoxic T-cell responses, thereby targeting PRAME-expressing tumor cells and PSMA-expressing tumor cells or tumor-associated neovasculature. A completed Phase 1 study in advanced solid malignancies reported acceptable tolerability and vaccine-induced antigen-specific T-cell responses, without RECIST partial or complete responses.
