Overview

MKC253 is an inhaled formulation of glucagon-like peptide-1 (GLP-1, 7-36 amide) adsorbed onto Technosphere microparticles for oral inhalation. It was designed for rapid delivery of GLP-1 directly to the lungs for absorption into the bloodstream. In clinical studies, inhaled GLP-1 was absorbed extremely quickly, with peak concentrations occurring within 3-5 minutes after inhalation, and levels returned to baseline within 30 minutes. The formulation produced a higher ratio of active-to-total GLP-1 (approximately 67%) compared to intravenous administration (20-33%). MKC253 induced insulin secretion within 6 minutes of inhalation and reduced blood glucose levels for approximately 20 minutes, without the typical nausea and vomiting associated with high GLP-1 levels.
