MKlp2 inhibitors are a class of small molecule antimitotic agents that target motor kinesin-like protein 2 (MKlp2), also known as KIF20A. MKlp2 is a kinesin-6 family motor protein primarily known for its role in cytokinesis, where it facilitates the transport of the chromosomal passenger complex to the spindle midzone. Research conducted at The Ohio State University has identified a novel role for MKlp2 in early mitosis, showing that its inhibition induces a spindle assembly checkpoint-mediated arrest. This mechanism makes MKlp2 an attractive therapeutic target for glioblastoma, as its inhibition significantly reduces the survival of patient-derived glioblastoma cell lines while potentially avoiding the neurotoxicity associated with traditional microtubule-targeting drugs.
