ML167 is a potent and selective small molecule inhibitor of Mixed Lineage Kinase 3 (MLK3), also known as MAP3K11. It was originally identified through the NIH Molecular Libraries Probe Production Centers Network (MLPCN) as a high-affinity probe for MLK3, demonstrating an IC50 of approximately 160 nM and high selectivity over other kinases in the MLK family and the broader kinome. MLK3 is a member of the MAP3K family that regulates the JNK and p38 MAPK signaling pathways, which are involved in stress response and apoptosis. In addition to its established role as an MLK3 inhibitor, computational virtual screening and structure-based pharmacophore modeling have identified ML167 as a potential inhibitor of the Epidermal Growth Factor Receptor (EGFR) and its clinically relevant mutations (such as T790M and L858R), suggesting its potential utility in the treatment of EGFR-driven malignancies like non-small cell lung cancer.
