Overview

ML171 (2-acetylphenothiazine) is a potent, selective, cell-active small molecule inhibitor of NADPH oxidase 1 (NOX1). It was identified by the Scripps Research Institute Molecular Screening Center as part of the NIH Molecular Libraries Program. ML171 potently blocks NOX1-dependent reactive oxygen species (ROS) generation with nanomolar potency (IC50 of approximately 129–250 nM) and exhibits high selectivity over other NOX isoforms (NOX2, NOX3, NOX4) and xanthine oxidase. It is widely used as a chemical probe in preclinical research to investigate the role of NOX1-derived ROS in various pathological processes, including cancer cell invasion, cardiovascular remodeling, inflammation, and diabetic cardiomyopathy.
