Overview

ML180 is a small molecule antagonist of the nuclear receptor Liver Receptor Homolog-1 (LRH-1, also known as NR5A2). LRH-1 is a member of the NR5A subfamily of nuclear receptors and plays a critical role in regulating steroidogenesis, bile acid homeostasis, and cell proliferation. In the context of oncology, LRH-1 has been identified as a driver of castration-resistant prostate cancer (CRPC) by promoting intratumoral androgen biosynthesis and directly activating the androgen receptor (AR) promoter. ML180 was identified through the NIH Molecular Libraries Program as a selective chemical probe to study LRH-1 function. Research indicates that ML180 can attenuate the viability of enzalutamide-resistant prostate cancer cells and potentially reverse resistance to anti-androgen therapies by inhibiting LRH-1-mediated AR signaling.
