ML324 is a cell-permeable, small molecule inhibitor of the lysine demethylase 4 (KDM4/JMJD2) family of histone modifying enzymes. It specifically inhibits KDM4 demethylases, which are responsible for removing H3K9 trimethylation marks from histones. ML324 demonstrates potent antiviral activity against herpes simplex virus (HSV), human cytomegalovirus (hCMV), and Cyprinid herpesvirus 3 (CyHV-3), primarily by inhibiting viral immediate early (IE) gene expression. It also shows antimalarial activity by selectively inhibiting late-stage Plasmodium falciparum gametocytes through inhibition of PfJmjC3, leading to altered histone methylation and aberrant gene expression in the malaria parasite[2][5][7]. Additionally, ML324 has been shown to reduce tumor volume and growth in preclinical cancer models[9].
