ML351 is a potent, selective, and reversible small molecule inhibitor of **12/15-lipoxygenase (12/15-LOX)**, also known as **Arachidonate 15-lipoxygenase (ALOX15)**. Developed through the NIH Molecular Libraries Program, it serves as a high-affinity chemical probe with significant selectivity over other lipoxygenase isoforms such as 5-LOX and 12-LOX. By blocking the enzymatic activity of 12/15-LOX, ML351 prevents the production of pro-inflammatory and pro-oxidant lipid mediators, thereby exerting neuroprotective effects. Preclinical research has demonstrated its ability to reduce neuroinflammation, mitigate hippocampal neuronal damage, and improve cognitive and behavioral outcomes in models of **status epilepticus**, **ischemic stroke**, and **traumatic brain injury**. It is currently utilized as a research tool to investigate the role of lipid signaling in neurodegeneration and oxidative stress-related pathologies.
