mLAG-3Ig is a recombinant fusion protein consisting of the extracellular domain of murine Lymphocyte Activation Gene-3 (LAG-3) fused to the Fc region of a murine IgG2a antibody. Developed by Ares Advanced Technology (a subsidiary of Serono, now part of Merck KGaA), it serves as a potent vaccine adjuvant. The protein functions by binding with high affinity to MHC class II molecules on professional antigen-presenting cells (APCs), such as dendritic cells. This engagement triggers APC activation and maturation, subsequently enhancing both humoral and cellular immune responses, specifically promoting Th1-type cytokine production and CD8+ T cell expansion. It has been primarily utilized in preclinical research to improve the immunogenicity of subunit vaccines against cancer and infectious diseases.
