MLINK Duo is a proprietary, cleavable peptide-based linker technology developed by Debiopharm as part of its MultiLINK™ ADC Technology Suite. It is specifically engineered to enable the simultaneous attachment of two distinct therapeutic payloads (warheads) to a single antibody, facilitating the coordinated delivery and release of multiple agents within cancer cells. The linker is designed to be cleaved by cathepsin B, a lysosomal enzyme, ensuring targeted release of the payloads. This technology supports high drug-to-antibody ratios (DAR), potentially reaching DAR 8+8, and is intended to overcome treatment resistance in tumors by maximizing synergistic therapeutic effects while minimizing systemic toxicity.
