MLL3 siRNA is a small interfering RNA designed to silence the expression of the MLL3 (Myeloid/Lymphoid or Mixed-Lineage Leukemia 3) gene, also known as KMT2C. MLL3 is a histone lysine methyltransferase that primarily catalyzes the monomethylation of histone H3 lysine 4 (H3K4me1) at enhancer regions, a key epigenetic modification for gene activation. In research contexts, particularly concerning rheumatoid arthritis (RA), MLL3 siRNA is used to investigate the role of MLL3 in the activation of synovial fibroblasts (RASFs). Studies have shown that siRNA-mediated depletion of MLL3 suppresses the expression of pro-inflammatory chemokines such as CCL5, CXCL10, and CXCL11, suggesting that MLL3 contributes to the inflammatory environment in RA joints through enhancer-mediated epigenetic regulation.
