MLN2704 is an investigational antibody-drug conjugate (ADC) developed for the treatment of metastatic castration-resistant prostate cancer. It consists of a humanized monoclonal antibody (MLN591) that targets prostate-specific membrane antigen (PSMA), which is highly expressed on prostate cancer cells, conjugated via a disulfide linker to the cytotoxic maytansinoid DM1, a potent microtubule inhibitor. Upon binding to PSMA-expressing tumor cells, MLN2704 is internalized and releases DM1 inside the cell, where it binds to tubulin and disrupts microtubule assembly, leading to cell death. Clinical trials showed limited efficacy and significant neurotoxicity due to instability of the linker and rapid deconjugation of DM1 from the antibody[1][2][4][5][6].
