Overview

MLN3126 is an orally available small molecule that acts as a potent and selective antagonist of the chemokine receptor C-C chemokine receptor 9 (CCR9). CCR9 is the homing receptor for C-C motif chemokine ligand 25 (CCL25), which plays a key role in mucosal immunity and the pathogenesis of inflammatory bowel disease (IBD) by recruiting T cells into the gut mucosa. By inhibiting CCR9, MLN3126 blocks CCL25-induced calcium mobilization and T cell chemotaxis, thereby reducing inflammation in preclinical models of colitis. The drug was developed as a potential treatment for IBD, particularly colonic forms such as ulcerative colitis. Preclinical studies demonstrated its efficacy in mouse models of T cell-mediated colitis[1][2][3][7]. Clinical development included early-phase trials to assess safety and pharmacokinetics[5].
