MLN8327 is a small molecule inhibitor of Polo-like kinase 1 (PLK1), a serine/threonine kinase that serves as a master regulator of the cell cycle, particularly during mitosis. Developed by Millennium Pharmaceuticals (a subsidiary of Takeda), MLN8327 selectively targets the kinase domain of PLK1, which is essential for centrosome maturation, bipolar spindle assembly, and chromosome segregation. In preclinical research, including studies on pediatric gliomas and glioblastoma (GBM), MLN8327 has demonstrated efficacy in inhibiting the growth of both cancer stem-like cells (neurospheres) and non-stem monolayer tumor cells. By disrupting mitotic progression, the compound induces G2/M phase arrest and subsequent apoptosis in rapidly dividing malignant cells. Its development has focused on high-grade malignancies where PLK1 is frequently overexpressed and associated with poor prognosis.
