Drug intelligence / Profile preview
MLS-0046818 is a novel small molecule negative allosteric modulator (NAM) that selectively targets the corticotropin-releasing factor-binding protein (CRFBP) and corticotropin-releasing hormone receptor 2 (CRHR2) complex. Developed jointly by Brown University and the Sanford Burnham Prebys Medical Discovery Institute, MLS-0046818 was identified through a high-throughput screen of over 350,000 compounds. It selectively inhibits the excitatory function of the CRFBP-CRF2 complex without affecting CRF1 receptor activity or CRF2 receptor function in the absence of CRFBP. MLS-0046818 is being investigated as a preclinical research tool and therapeutic candidate for the treatment of stress-related neuropsychiatric conditions, specifically substance use disorders (SUD) and alcohol use disorder (AUD).
