MLS-269 is a first-in-class small molecule inhibitor of the tetraspanin protein CD151, currently under development by MLS Bio (formerly Esfam Biotech). It specifically targets the CD151-integrin interface to disrupt signaling pathways essential for tumor cell motility, survival, and vascular invasion. Unlike traditional cytotoxic agents, MLS-269 employs a host-directed approach to reduce tumor invasiveness and metastatic dissemination without directly inhibiting tumor proliferation, which may help mitigate the development of drug resistance. The drug is primarily being developed for metastatic castration-resistant prostate cancer (mCRPC) and non-metastatic castration-resistant prostate cancer (nmCRPC), with potential future applications in other CD151-high malignancies such as kidney, lung, pancreatic, and breast cancers.
