MM-003 (also known as Gal-3C or Galectin-3C) is a recombinant protein-based biologic drug candidate developed by MandalMed, Inc. It functions as a dominant-negative inhibitor of galectin-3, a protein that is a key mediator of organ fibrosis and harmful tissue remodeling. The drug is engineered from the C-terminal carbohydrate recognition domain (CRD) of human galectin-3, which allows it to bind to galectin-3 ligands and block the activity of endogenous galectin-3 without triggering pro-fibrotic signaling pathways. MM-003 was primarily developed to prevent and treat harmful cardiac remodeling and fibrosis following myocardial infarction (heart attack) to improve cardiac function and reduce mortality from subsequent heart failure. It was also investigated for the treatment of liver fibrosis and cirrhosis. Preclinical studies in animal models demonstrated that MM-003 could limit excessive fibrosis and showed superior therapeutic efficacy compared to standard treatments like losartan and spironolactone. However, according to market reports, the development of MM-003 was abandoned by MandalMed as of 2023.
