Overview

MM-111 + lapatinib + trastuzumab is an investigational combination therapy targeting HER2-positive cancers. This regimen consists of: - **MM-111**, a bispecific antibody fusion protein targeting HER2 (ERBB2) and HER3 (ERBB3), designed to inhibit the HER2-HER3 signaling axis by binding to both receptors and forming a receptor/MM-111 complex, preventing HER2-HER3 heterodimerization and thus suppressing downstream PI3K/AKT pathway signaling. - **Lapatinib**, a reversible small molecule tyrosine kinase inhibitor targeting EGFR (ERBB1) and HER2 (ERBB2), blocking their kinase activity and downstream signaling. - **Trastuzumab**, a monoclonal antibody binding to HER2 (ERBB2), prevents HER2-mediated signal transduction, promotes antibody-dependent cell-mediated cytotoxicity (ADCC), and blocks downstream survival signaling. The combination is under investigation for overcoming resistance seen with HER2-targeted therapies and for maximizing growth inhibition or inducing cancer cell death in HER2-overexpressing tumors[2][3][4].
