Overview

MM-111 + trastuzumab is a combination therapy under investigation for the treatment of HER2-positive cancers, particularly breast and gastroesophageal (GE) cancers. MM-111 is a novel bispecific antibody fusion protein that specifically targets the HER2/HER3 heterodimer, blocking heregulin-induced activation of HER3 and downstream signaling pathways associated with tumor growth and resistance to anti-HER2 therapies. Trastuzumab is a humanized monoclonal antibody that binds to the extracellular domain of Human epidermal growth factor receptor 2 (HER2), inhibiting proliferation and survival of cancer cells overexpressing this receptor. The combination aims to overcome resistance mechanisms mediated by HER3 activation in tumors treated with anti-HER2 agents like trastuzumab. Clinical trials have evaluated this combination in advanced breast cancer and GE junction/gastric adenocarcinoma[5][6][8].
