Overview

MM-141 + everolimus is an investigational combination therapy consisting of MM-141, a bispecific tetravalent monoclonal antibody that targets and antagonizes both IGF-1R (Insulin-like growth factor 1 receptor) and ErbB3 (HER3), and everolimus, a small molecule inhibitor of the mTOR (mechanistic target of rapamycin) pathway. This combination aims to overcome resistance mechanisms in cancers where IGF-1R and ErbB3 co-activation drive tumor growth and adaptive signaling, particularly in solid tumors. MM-141 blocks and promotes the degradation of both IGF-1R and ErbB3, preventing compensatory survival signaling often seen in cancer cells, while everolimus directly inhibits mTORC1, thereby blocking downstream PI3K/Akt/mTOR signaling. The combination has been evaluated in Phase 1 studies for advanced solid tumors, primarily to assess safety, tolerability, and pharmacological effects[1][2][3].
