mm-miR-378a-3p mimic is a synthetic double-stranded RNA designed to replicate the biological activity of the endogenous mouse microRNA miR-378a-3p. In preclinical research, specifically in murine models of intestinal fibrosis (such as the DSS-induced model), the administration of this mimic has been shown to exacerbate chronic inflammation and fibrotic progression. Despite miR-378a-3p levels being naturally reduced in fibrotic colonic tissue, exogenous replacement via intravenous injection leads to increased neutrophil infiltration, architectural distortion of the glandular epithelia, and elevated expression of pro-fibrotic and pro-inflammatory markers, including Tgfb1, Il1b, and Mmp2. In human intestinal fibroblasts, the human ortholog (hsa-miR-378a-3p) has been observed to upregulate the anti-apoptotic molecules BCL2 and MCL1, potentially contributing to fibroblast persistence in chronic disease states.
