**MMAE conjugated IgG-F1** is a preclinical, human IgG1-based antibody-drug conjugate directed against Nectin-2, a cell-surface adhesion molecule and immune regulator overexpressed in a substantial proportion of breast cancers. The construct uses the anti-Nectin-2 antibody IgG-F1, a rapidly internalizing antibody with reported 1 nM affinity, coupled through an unspecified cleavable linker to the antimitotic payload monomethyl auristatin E. Following Nectin-2-mediated internalization and intracellular payload release, MMAE disrupts tubulin-dependent microtubule dynamics to produce tumor-cell cytotoxicity. In breast cancer cell lines, the construct showed reported IC50 values of 1.24-11.76 nM and induced dose-dependent tumor regression in an MDA-MB-468 triple-negative breast cancer xenograft model.
