A monoclonal antibody targeting matrix metalloproteinase-14 (MMP-14), also known as membrane-type 1 matrix metalloproteinase (MT1-MMP). MMP-14 is a key enzyme involved in the degradation of the extracellular matrix, a process essential for tumor cell invasion, metastasis, and angiogenesis. It also functions by activating other proteases, such as pro-MMP-2, and modulating angiogenic signaling pathways. The most prominent candidate in this class is **DX-2400**, a fully human IgG4 monoclonal antibody discovered using phage display technology. DX-2400 selectively inhibits the catalytic activity of MMP-14 without affecting other matrix metalloproteinases. Preclinical studies have demonstrated that this antibody can disrupt tumor-stromal communication, suppress the growth of primary tumors, and inhibit metastatic spread, particularly in models of advanced ovarian cancer. Its efficacy in reducing tumor vascularization has been shown to be comparable to established anti-angiogenic agents like bevacizumab.
