Drug intelligence / Profile preview
MMP-9 blocking antibodies are monoclonal antibodies designed to selectively bind and neutralize matrix metalloproteinase-9 (MMP-9), a zinc-dependent endopeptidase involved in the degradation of extracellular matrix components, particularly type IV collagen in the basement membrane. MMP-9 is rapidly upregulated following acute brain injuries, such as ischemic stroke and intracerebral hemorrhage (ICH), where it contributes to blood-brain barrier (BBB) disruption, neuroinflammation, and hemorrhagic transformation. By selectively targeting vascular MMP-9 without crossing the blood-brain barrier, these blocking antibodies can prevent BBB breakdown, reduce brain swelling, and limit hemorrhage volume and neurobehavioral disability. Preclinical research, including studies from the University of Arizona and Translational Sciences, Inc., has demonstrated that intravascular administration of specific MMP-9 blocking antibodies significantly improves outcomes in experimental models of stroke and ICH.
