Overview

**MMV055** is a novel antimalarial compound in early clinical development, designed as a long-acting injectable for malaria prevention. It acts by inhibiting the *cytochrome bc1 complex (Qi site)* of malaria parasites, which disrupts mitochondrial electron transport essential for parasite survival. MMV055 is structurally distinct from atovaquone, binding to a different site on the same target, and shows no cross-resistance with atovaquone-resistant parasite strains. Its activity targets malaria parasites in the liver, bloodstream, and even in the mosquito vector, making it suitable for both treatment and prevention. MMV055 is a potential partner for MMV371 in fixed-dose combinations aimed at extended malaria prophylaxis. The compound is being developed as a long-acting injectable (LAI) and is expected to enter first-in-human studies in 2025[1][3][5][7][8].
