Overview

MMV172 is a small molecule antimalarial compound identified through the Medicines for Malaria Venture (MMV) Malaria Box initiative. It belongs to the aminopyrimidine chemical class and exhibits potent activity against the erythrocytic stages of *Plasmodium falciparum*. The primary mechanism of action for MMV172 is the inhibition of the parasite's P-type ATPase 4 (PfATP4), a sodium-exporting pump located on the plasma membrane that is essential for maintaining low intracellular sodium concentrations. Inhibition of PfATP4 leads to a lethal disruption of sodium homeostasis, resulting in rapid parasite swelling and lysis. This mechanism is shared with several clinical-stage antimalarial candidates, such as cipargamin (KAE609). MMV172 serves as a valuable chemical probe for studying parasite biology and as a starting point for lead optimization in the development of next-generation antimalarial therapies.
