Overview

MMV367 is a synthetic pyrrolidinamide antimalarial clinical candidate developed for the treatment of uncomplicated malaria. It demonstrates rapid activity against *Plasmodium falciparum* and has no cross-resistance to current antimalarial drugs. The compound acts by interfering with the function of parasite acyl coenzyme A synthetase 10/11, an essential enzyme in asexual blood stage parasites. First-in-human studies have shown that MMV367 is orally bioavailable and has an acceptable safety and tolerability profile in healthy volunteers. Pharmacokinetic data indicate a half-life of approximately 16–19 hours and increased exposure when administered with food. The drug is being developed for both adults and children as a potential new therapy for malaria. Developed by Medicines for Malaria Venture and GSK.
