Drug intelligence / Profile preview
MMV665924 is a small molecule antimalarial compound identified from the Medicines for Malaria Venture (MMV) Malaria Box. It acts as an inhibitor of *Plasmodium falciparum* acyl-CoA synthetase 10 (PfACS10), an essential enzyme involved in activating scavenged fatty acids for lipid biosynthesis. Inhibition of PfACS10 by MMV665924 disrupts triglyceride formation, leading to a reduction in triacylglycerols and a buildup of lipid precursors, which ultimately kills the asexual blood-stage parasites. Resistance to MMV665924 is mediated by mutations in PfACS10 as well as mutations in the non-essential acyl-CoA synthetase 11 (PfACS11), which acts as a resistance mediator.
