Overview

MMZC-DM4 is a first-in-class DNA-Zip Code Drug Conjugate (ZDC) being developed by Kodikaz Therapeutic Solutions for the treatment of multiple myeloma. The therapeutic consists of a specific DNA sequence, termed a "Zip Code" (specifically MMZC1, derived from the AluSp retrotransposon), conjugated to the cytotoxic payload DM4, a potent microtubule inhibitor. This technology leverages the natural tropism of circulating tumor DNA (ctDNA) to achieve cell-specific targeting. MMZC-DM4 is designed to selectively bind and enter multiple myeloma cells, where the DM4 payload disrupts microtubule assembly, leading to cell cycle arrest and apoptosis. Preclinical data presented at ASH 2023 demonstrated that MMZC-DM4 has a potent killing effect on multiple myeloma cell lines in vitro and significantly reduces tumor growth in xenograft models with minimal systemic toxicity.
