Mn-PEGCG is a nanoparticle-based therapeutic composed of a metal-polyphenol network formed by the polymerization of epigallocatechin gallate (EGCG) and subsequent complexation with manganese ions (Mn2+). The resulting Mn-PEGCG nanoparticles are designed for oral administration and target sites of inflammation in the colon, particularly for the treatment of ulcerative colitis. Mechanistically, these nanoparticles act as antioxidants by scavenging reactive oxygen species (ROS), reducing pro-inflammatory cytokine production, and protecting colon cells from oxidative stress. Additionally, EGCG within the formulation promotes tight junction protein expression and enhances intestinal epithelial barrier function. In preclinical mouse models of ulcerative colitis, oral administration led to reduced intestinal inflammation, improved mucosal structure, increased mucus secretion, and strengthened intestinal barrier integrity[1][2].
