Overview

MNC2 (also known as huMNC2) is a humanized monoclonal antibody developed by Minerva Biotechnologies that specifically targets MUC1* (mucin 1 star), the tumor-associated, cleaved growth factor receptor form of MUC1. Unlike full-length MUC1, which is widely expressed on normal epithelial tissues, MUC1* is selectively exposed on cancer cells after cleavage and shedding of the tandem repeat domain. MNC2 binds to a conformational epitope on MUC1* that is unmasked in the tumor microenvironment, preventing the binding of its activating ligand, NME7. MNC2 has been utilized as the antigen-binding single-chain variable fragment (scFv) in chimeric antigen receptor (CAR) T-cell therapies, such as huMNC2-CAR44 and huMNC2-CAR22, which are in clinical development for metastatic breast cancer. Additionally, MNC2 is being investigated in preclinical studies as an antibody-drug conjugate (ADC), such as MNC2-deruxtecan, for the treatment of solid tumors.
