Overview

MNC2-MMAF is an antibody-drug conjugate (ADC) consisting of the humanized IgG1 monoclonal antibody huMNC2 conjugated to the cytotoxic payload monomethyl auristatin F (MMAF). The antibody component selectively targets MUC1*, a growth factor receptor formed by the tumor-associated enzymatic cleavage of the N-terminal portion of MUC1. MUC1* is expressed on over 75% of solid tumors but is absent on normal tissues. Upon binding and internalization, the MMAF payload, an antitubulin agent, inhibits cell division by blocking tubulin polymerization, leading to cell death. Developed by Minerva Biotechnologies, MNC2-MMAF has shown preclinical efficacy in various solid tumor models, including breast and pancreatic cancers, with no observed off-tumor toxicity in animal studies.
