mNEXSPIKE is a next-generation messenger RNA (mRNA) vaccine developed by Moderna for active immunization to prevent coronavirus disease 2019 (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). It is specifically indicated for individuals who have previously received any COVID-19 vaccine and are either 65 years of age or older, or between 12 and 64 years old with at least one underlying condition that puts them at high risk for severe outcomes from COVID-19. The vaccine contains nucleoside-modified mRNA encoding the N-terminal domain (NTD) and receptor-binding domain (RBD) of the Spike glycoprotein of the SARS-CoV-2 Omicron variant lineage JN.1, delivered in a lipid nanoparticle formulation. Clinical trials demonstrated noninferior—and in some subgroups superior—immunogenicity and efficacy compared to Moderna’s original Spikevax, with a similar safety profile but lower dose per injection[1][4][5][7][10].
