MNK-010 is a liposomal formulation of a Toll-like receptor 7 (TLR7) agonist developed by Mallinckrodt Pharmaceuticals. As an immunotherapy, it was designed to activate the innate immune system, specifically targeting TLR7 to induce the production of cytokines and enhance the antitumor immune response within the tumor microenvironment. The liposomal delivery system was intended to optimize the drug's distribution and minimize systemic side effects associated with TLR7 activation. MNK-010 entered a Phase 1 first-in-human dose-escalation study for patients with advanced solid malignancies who had exhausted standard treatment options; however, Mallinckrodt terminated the clinical program in 2018 for strategic reasons.
