MNP-EF2K siRNA is a nanoparticle-formulated gene therapy consisting of magnetic nanoparticles (MNPs) incorporating small interfering RNA (siRNA) targeting eukaryotic translation elongation factor 2 kinase (eEF-2K) [1125]. Developed by researchers at the University of Texas MD Anderson Cancer Center and Middle East Technical University (METU), eEF-2K is an atypical kinase that is highly upregulated in BRCA1-mutated breast cancers, where it promotes cell survival, proliferation, and drug resistance [1125]. By silencing eEF-2K expression, MNP-EF2K siRNA inhibits tumor cell proliferation, angiogenesis, and induces apoptosis [1125]. In preclinical models of BRCA1-mutated breast cancer, systemic administration of MNP-EF2K siRNA significantly inhibits tumor growth and sensitizes cancer cells to PARP inhibitors such as olaparib [1125].
