MNP-PL is a novel nanoparticle formulation consisting of the natural naphthoquinone plumbagin (PL) encapsulated within magnetic nanoparticles (MNPs) featuring an iron oxide core. Developed by researchers at the University of Tennessee Health Science Center, MNP-PL is designed to treat pancreatic cancer by improving the bioavailability and reducing the systemic toxicity of plumbagin. The formulation targets the desmoplastic microenvironment of pancreatic tumors, specifically inhibiting the oncogenic CXCL12/CXCR4 signaling axis. In preclinical studies, MNP-PL demonstrated superior growth inhibition and suppression of colony formation in human pancreatic cancer cells compared to free plumbagin, while also decreasing the expression of CXCR4 and its ligand CXCL12 (SDF-1) in both cancer and stromal fibroblast cells.
