Overview

MNPR-101-RIT is a novel, first-in-class radio-immuno-therapeutic designed for the treatment of advanced solid tumors that express the urokinase plasminogen activator receptor (uPAR). It consists of the humanized monoclonal antibody MNPR-101 conjugated to therapeutic radioisotopes, including lutetium-177 (Lu‑177) and actinium‑225 (Ac‑225), enabling targeted delivery of cytotoxic radiation to uPAR-expressing cancer cells. Preclinical studies have demonstrated compelling and durable anti-tumor activity, with near-complete elimination of uPAR-positive tumors after a single injection in animal models. The primary indications under development include triple-negative breast cancer, colorectal cancer, pancreatic cancer, bladder cancer, and ovarian cancer. Monopar Therapeutics is advancing this agent as a precision radiopharmaceutical therapy for cancers with high unmet need[1][5][6][10].
