Mocaciclib (Q901) is a highly selective, covalently binding small molecule inhibitor of cyclin-dependent kinase 7 (CDK7) being developed by Qurient. CDK7 is a critical regulator of both the cell cycle (as part of the CDK-activating kinase complex) and gene transcription (as part of the TFIIH complex). By targeting CDK7, mocaciclib disrupts the progression of the cell cycle and impairs DNA damage repair mechanisms, leading to selective apoptosis in cancer cells. The drug is currently undergoing Phase 1/2 clinical evaluation for the treatment of various advanced solid tumors, including ovarian, breast, and small-cell lung cancers, and is being studied both as a monotherapy and in combination with immune checkpoint inhibitors and antibody-drug conjugates.
