Overview

MOD-6030 is a long-acting recombinant fusion protein consisting of a glucagon-like peptide-1 (GLP-1) analog covalently linked to alpha-1 antitrypsin (AAT). Developed by OPKO Health (following its acquisition of Prolor Biotech), this therapeutic candidate is designed for the treatment of type 2 diabetes and potentially type 1 diabetes. The drug leverages a dual mechanism: the GLP-1 moiety acts as an agonist at the GLP-1 receptor to stimulate glucose-dependent insulin secretion and suppress glucagon release, while the AAT moiety provides potent anti-inflammatory and protease-inhibitory effects. AAT is intended to protect pancreatic beta cells from inflammatory damage and apoptosis, potentially preserving islet function in both autoimmune and metabolic contexts. Furthermore, the large molecular size of AAT serves as a half-life extender for the GLP-1 peptide, significantly reducing the frequency of subcutaneous administration compared to native GLP-1.
